If use cassian's 2 pip "Last Stand" to issue an order to R2D2 who has comms relay, and I use the relay to give the order to Luke. Does Luke get the benefits of the card or not?

No, in that case R2 will be the one issuing the order to Luke, not Cassian, so the requirement of Last Stand ("When Cassian Andor issues an order...") is not fulfilled.
