If a unit tries to go up the side of a building with a ladder, do they still have to role to see if any die?

No, they dont, as long as you and your opponent have agreed in advance what walls the clamber rules dont apply to. Ie a wall with a ladder or stairway or something similar. Its not an automatic rule for the ladder itself, but a ladder can indicate to you and your opponent what terrain is climable without penalty.
