This COTDP Mythos card allows the first player to choose one investigator to become Blessed & another investigator to become Cursed. Can he pick an investigator who is already Cursed --- thereby nullifying the negative effect of this card?

I don't see anything against this

Note that there is one small effect: the old Curse replaces the new one, so that investigator will not be allowed to roll for it on the following Upkeep.

Tibs speaks truth. And, to be honest, that's a much better option than 2 investigators cursed.
