If for example Cersei is attacking in an intrigue ( she does not kneel ) can this location work ?

My feeling is yes because it does not state THEN remove from the challenge .

You're correct. It also doesn't target kneeling characters so it'll work that way for sure.

It's the same with the way Frozen Outpost works. You can target any defending character and they will get the +2 STR even if they were already standing.
