During the standing phase in a game of melee do you also stand titles ?

Is the plot card you revealed still active until you reveal the next plot card ? If so…
Could i for example, bring out 'Shadow Politics' from shadows prior to revealing a new plot card and then in theory if i had 'Focussed Offence' as my plot card could i then initiate an intrigue ( as per Shadow Politics ) and get a claim of three ?
Technically you do not kneel Titles to activate or use their effects. Titles that become "spent" or "used" can only be used one time per round. It's common practice, however, to kneel titles to represent that you have already used them(like Hand of the King for the 2 influence). However, standing them up during the standing phase does not allow you to spend 2 more influence before you return it at the end of the round.
And to answer your question about plots, they are still your revealed plot until you reveal a new one. If Focused Offense is revealed for you and you have already declared one challenge, you won't be able to declare another challenge until you reveal a new plot. This includes during the plot phase before you reveal your next plot.

What makes you think that a revealed plot card is ever inactive? Revealed plot text is always active unless it is blanked.
It's not "revealed until the end of a round", otherwise the rules would tell you this. Until you reveal a new plot card, you are following the text on your currently revealed plot.
Plots are in 3 states: Used, Revealed, and Plot Deck.
Focused Offense's restriction of only allowing you to make a single challenge would still be active. If you've made a challenge already in the previous turn, the plot's restriction is still active and you cannot make any more challenges. If you for some reason had not made a challenge during the previous turn then yes, you could do what you describe.
The restriction of only making a single challenge will always apply pre-plot even though it's technically a new turn - the plot's restriction does not turn off until it stops being your revealed plot.

Of course - as I type this - I now think of an odd situation. What if someone pins this plot with Art of Seduction and you've already made the challenge? Does the constant effect apply and you are now unable to make any challenges on the next turn since your plot didn't change? If not, why not?

That's exactly what will happen. Focused Offense doesn't provide you a duration of the effect, so it must be as long as the plot is revealed.
If it said "When revealed, ….. this challenge phase.", then it would only be restricted to the upcoming challenge phase of that round. It would also stick with the effect even if a new plot was revealed because it would be creating a lasting effect.
Without "when reveal … [duration].", plot text is only active until a new plot is revealed.
Wow - that would suck. Glad I don't ever play that plot. LOL! :-D
Also note, to hi-jack the thread a little more, that if you get stuck (or stick your opponent) with Fear of Winter that will be two entire turns you can only play 1 card total. Similar to Focused Offense, FoW does not have a duration of it's effect. It simply IS in effect until the plot is no longer revealed.
Actually, FoW has an explicit duration of "until you reveal a new plot card". This happens to be redundant, because that's the duration it would have anyway, but it's still written on the card.
