A basic quesion, but I'm new to the game, and I don't see any clarification in the ruels or FAQs.
When calculating Dominance pts, do you use the unmodified strength of standing characters, or do you apply any modifiers from locations etc.

You apply all modifications. The game uses the term "printed STR" for the unmodified strength, and unelss that term is used it is always the modified strength which matters.
Or, said another way, when you are told to look at a card's STR, it always means the current, "as modified" STR. If you look at anything other than the current, "as modified" STR, the card will tell you that specifically.

The most common way to differentiate between modified and unmodified costs, strengths, etc. is to look for the word "printed". When a card refers to "printed strength" or "printed cost", that means the unmodified strength or cost. I can't think of an example of a card that doesn't use this terminology yet does refer to unmodified strength or cost, but one may exist.
