You need a target lock but the card does not say "spend your target lock to use this card." So you can still have it to reroll your misses. Use push the limit and have a focus as well…… Poof!

Homing Missiles against 4 agility dice + Focus is a 50/50 shot. If Tycho can Target Lock and Focus in the same round, the likelihood of stripping Focus goes up to 75/25. For Wedge, it's about 43/57. But I think that's about as heavily as you can lean on the odds, unless you go the opposite direction and just try to overwhelm them with weak attacks.
[EDIT: Forgot to subtract an agility die for Wedge's ability! It's actually more like 65/35, which is pretty good odds considering he can do it every round.]
I'm trying to think of what to do on the Imperial side. Krassis + Heavy Laser Cannon makes sense, but I'm having a hard time settling on what to match him with…
I'm thinking for the Empire, Interceptors at range 1, but with Boost on the Stealthed Awings, getting shots on them at Range 1 is not an easy proposition.

To reiterate: Homing Missiles DOES require a target lock to use, but does NOT require spending said lock. Look at the wording:
Concussion Missiles: ATTACK [Target lock]: Spend your target lock and discard this card to perform this attack.
Homing Missiles: ATTACK [Target lock]: Discard this card to perform this attack.
So with the Homing Missiles the target lock is available to reroll attack dice results.

could always get another TIE to come in and block the target, prevent him from boosting. Of course, that does mean that particular TIE will get range 1-ed as well
Yes, I've read the card now again (really don't understand how I missed it in the first place) and as it has been pointed out, I didn't read the whole card, just assumed it to be like the rest of the missile upgrades. My bad.
So that makes the Homing missiles really good at picking of empire ships who rely on the evade tokens. And yes, they do work great on other ships aswell when getting the chanse to re-roll the damage it does. Nice.
