The answer is "no".

"Any Phase: Kneel Old Nan to choose a character. Until the end of the phase, that character gains or loses a Trait of your choice."

Lord and Lady are TWO separate Traits -- "Lord" & "Lady", not "Lord and Lady". You can kneel Old Nan to make a character gain "Lord" or "Lady", but not both.

"When revealed, the opponent to your left chooses 1 Lord and 1 Lady character, if able. Then, you must choose and kill 1 of those characters. The other claims 2 power."

First, your opponent (not you) chooses two characters (one Lord and one Lady). Then you pick one of those characters and kill it. The character that was NOT killed claims 2 power.

But if my opponent have only one lord and i add the "lady" trait to that lord, and its the only lord+lady?

The plot specifies 1 Lord and 1 Lady. And then specifically says 1 dies and the other claims 2 power. So you can't choose just one card for the effect nor can you kill and give the power to the same character.

The most straightforward answer is that you cannot choose the same card as the target of a single effect more than once. Any time an effect has 2 or more targets, choosing card X as target #1 makes it illegal to choose the same card X as target #2.

The clearest example of this is that if you lost a 2-claim military challenge and choose Eddard Stark for claim target #1, it is illegal to also choose that same copy of Eddard Stark for claim target #2 - even if you could save him as claim target #1. So, in the same way, if you choose Eddard as the "Lord" target for Red Wedding, that makes it illegal to choose him as the "Lady" target, even if he has gained the Lady trait.
