If my opponent has Tourney for the hand as active plot, can his knights without crests defend against Wex Pyke?

Yes. Wex resolves on characters, changing their eligibility to be declared as defenders. Immunity would allow those characters to ignore the change in their ability - and thus they can be declared as defenders (assuming the right icons, etc.)

shocked at that answer actually. not what i expected to hear. second time today too (baelor/shadow event thread).

I think people get thrown by things like this because they parse it differently than it's actually written.

They see it as something like "Oh, since Wex's attacking I can't declare blockers without crests." Instead of "Wex makes those characters unable to block"

The two work the same almost all the time, but when immunity's involved the wording becomes crucial.

There are a lot of examples in the game where the exact phrasing makes no real difference in outcome, but all the difference in the world for immunity.
