I'm not sure what you mean by cancelling the challenge. The challenge itself is way too far along to cancel. If you asking about canceling the determination of winner again, it wouldn't need to open a new window, that window would still be open as long as players do not pass on the option to play save/cancel responses.

So player 1 could play Misinformation, then player 2 could play Burning on the Sand, if there's a player 3, they could play another card, etc.

To simplify, you need to play it between determining who won the challenge and resolving the claim effect.

Yes. Your opponent can then play their own Misinformation or Burning in the Sands to cancel you as that challenge winner directly after that.
