If I have three copies of Castellan in play, am I still only allowed to activate its response once overall? Or can I trigger it once for each copy of the character in play?

One Limited Response per round, so you only get to activate one Castellan. That also means you couldn't activate any other Limited Responses (like, say, the Alchemist's Guild Hall) for the remainder of the round.
