River Blockade is in play.
The opponent has in play Eastern Fiefdoms and Khal Drogo's Tent. The opponents wants to play Flame-Kissed with ambush.
a) Does River Blockade cancel the influence of one location? And if yes who chooses.
b) The Flame-Kissed remains in the hand of the player or is discarded?
And if discarded what happens if a player wants to play a character. Is he discarded? Happens something else?
I believe kneeling influence to pay a cost isn't a card effect (though you'd better wait for confirmation from someone more knowledgeable, like Ktom).
Assuming River Blockade prevents one of the location from providing influence, if the Targaryen player has no other sources of influence, he cannot pay the cost to ambush Flame Kissed, so he cannot even attempt to put it into play and it stays in his hand.
From the FAQ:
Keyword effects with a point of initiation
(such as ambush) cannot be canceled.
Ambush is the triggered effect that requires the influence to be knelt as a cost, and could be canceled by River Blockade if the FAQ didn't preclude it. Kneeling an influence isn't a triggered effect, and indeed, you can never just "decide" to kneel an influence - you can only do when another card instructs you to.
