I open the door (level 1) to set up the map of level 2. next, do I have to ignore the tiles and all monster of level 1? Do they follow me as I draw AI cards?

If the levels are connected (as they are in most missions, Scattered being the exception), then you have Locusts on lvl 1, lvl 2 and lvl 3, and ALL matching the AI card will activate.
