Does Sister Mary's fulfilled personal story (she cannot be cursed among other things) affect loss of blessing?
Basically she becomes perma-blessed if she completes her story. Some card effects state that a player is "cursed." So if she is in a state of being blessed, and she "can't be cursed," does that mean that a curse can't be applied to make her lose her blessing? (Hope that makes sense.)

I would say that the "can't be cursed" portion of her completed back story refers to the act of giving the Cursed card to Sister Mary. Basically, she can't have a Curse applied to her and therefore cannot lose her Blessing.

Be careful with this wording. If she can't be cursed, then she can't receive a Curse card. This means that a Curse card cannot nullify her Blessing, but there's no reason why she cannot lose her Blessing via the upkeep roll, or even some other rare effect that says "discard your blessing card" such as the revised Dark Phararoh's "Tainted" ability.

Interesting. i would have argued (and played it that way until now), that she can never have a Curse card, but that she can lose her blessing by being cursed.
Reason: "Cannot be cursed" can be interpreted as "can never be in the status: Cursed", but this would not prevent her from going from Blessed to normal.
However, if you read her PS "cannot be cursed" as "is never affected by an encounter, card or whatever "cursing" her, then her Blessing would remain.
I think both interpretations are valid and this is - one of many - FAQ entries ;-)

Here's the thing: an encounter would say "you are cursed ." If she can't be cursed, then it stands to reason that she can ignore this text. But we know that this text would normally lead to a blessed character canceling out the blessing, so it must be the case that she can't lose her blessing by gaining a curse card.
I do agree though, "are cursed" is an easily confusing choice of words.
It's more like you're saying "a curse cannot be applied to her."
"Sister Mary cannot receive a Curse card."
