Lets say a lord charcter who has upon him the Bodyguard card and a duplicate is the defender.

If there is a claim of 2can the defender discard the duplicate and the Bodyguard or he has to discard one of the 2 and kill another one charcter?

No. The other player must choose 2 different characters. Saving the character comes later and is not relevant when settling claim.
