Assuming I'm playing with the Brotherhood agenda but have no Brotherhood characters in play. If I win a power challenge, I understand I can't claim a power since I have no characters to put the power on but does my opponent lose a power?

But don't confuse the typical mechanics with what happens during the game. If you beat me in a power challenge, I don't discard a power from my House and you claim one for yours. The result is that power is moved from my House to yours (or, in the case of the Brotherhood Agenda, to one of your characters). If you have no Brotherhood characters, there is nowhere to move the power to. Therefor, the power never actually moves from my House card.
So if you are playing the Brotherhood Agenda and you have no Brotherhood cards in play, you do not gain power and your opponent does not lose power.
