As the title says, when does "While a friendly ship at range 0-2 defends..." actually happen? After the Attacker rolls his dice? Or can the Defender roll his dice before Luminara decides to play with the Attack results? Or does it happen during the attack resolution phase?

It would happen in the dice modification step. The attacker would roll their dice, then you could have the chance to use Luminara's ability. The attacker would then be allowed to apply any of their modifiers (rerolls, spending focus tokens, etc).

How would it work with say, Seyn Marana? Can you change the Crit result to a Hit before she decides to spend it?

Spending a result is still a modification, so Seyn would have to wait for Luminara to user her ability (if she wanted) before Seyn could spend the die result, since Defender modifies attack dice first.
The sequence:

Attacker Rolls Red Attack Dice
- Defender Modifies Attacker's Red Dice (Luminara)
- Attacker Modifies Attackers Red Dice (Focus/Lock/Calculate, Seyn, etc etc)

Defender Rolls Green Defense Dice
- Attacker Modifies Defender's Green Dice (Juke, Zuckuss)
- Defender Modifies Defender's Green Dice (Focus/Evade, Serissu, etc etc)
The easiest way to remember the order of dice mods is the owner of the dice modify them last.
With the exception of crackshot, because it occurs after the neutralise results step.

Crack Shot is also not technically a dice modification
The actual exception is Reinforce.
