If Mark uses the ability on Sophie “take 1 direct damage and receive +2 to the skill value for that test” does that trigger the ability on his character card?

Yup. Marks investigator card will trigger the ability when he takes damage.

Which is why you pack damage healing when you play Mark, to stay on Sophie's bad side.
