If I already have a copy of Darkstar in play and I mill a copy of Darkstar from my deck and put him into play as a duplicate?

Yes. You can use a "put into play" effect on a unique card you already have in play to have the new copy enter play as a dupe.

Note: If there is a copy in your dead pile, you can't put any of it into play.
