And as a rules clarification, when is "Defending" as her card states "when defending"? Is this as soon as you are designated the target of an attack or when you are physically throwing defense dice in the defense phase?

As soon as you are designated the target of an attack, you are defending.

As for when L3-37 triggers, its step 2:

1. Attacker rolls attack dice

2. Defender modifies attack dice

3. Attacker modifies attack dice

4... (and so on)

Also note, that the attacker cannot reroll his dice again during step 3.
