Seppun Ishikawa refers to cards with the Imperial trait (as shown by the bold italics). The Imperial Favor doesn't have any trait, so it doesn't count.
Imperial Favor isn't a card in play and had no traits. So the above is correct, No it doesn't work with Seppun
Other Question regarding the favor,

No, because discarding the favor is a cost of this action. You can't ignore stuff before a dash.
