If the other player attempts a mission in a system where i already have a leader stationed, is his mission then automatically opposed by my leader?

As long as it is an attempt mission and not a resolve mission, yes it does.

You can even oppose with a leader that has no icons in the particular skill. The person going on the mission just needs to roll 1 success, since by definition you roll zero.

Also, even if you already have a leader in that system, you could send another one and so both will oppose the mission
