Is it intended that the Spice tokens are removable once placed on the Rebel or Imperial drop-off points?

Neutral mission tokens can be retrieved and dropped by figure carrying one on either stockpile, they do not disappear.
So, during the mission any figure (except ones with the Non-Sentient ability) can retrieve tokens from either stockpile. I.e. your stockpile will probably need some guarding if you intend to keep yours.
Note that because the retrieve and drop are performed without requiring actions, a figure can share a space with another figure to perform them as long as they have enough movement points to get into a valid space to end movement afterwards.

I like the mission, btw. It allows a lot of different strategies (including stealing), and is thus very replayable.
Do you need to take 3 dmg in one hit to drop the barrel? Or in one characters turn? Or over time while carrying the spice? At that point in the story we found it very difficult to do 3 dmg with one attack. Meaning that the only way to have an imperial figure drop one was to kill them.
The imperials never picked one up until the last mission when he has more activations than us and he spent the last mission having all of his storm troopers and the SPOILERS(hunters that spawn right next to the rebel stock pile) take all of the barrels off of our stockpile in the last turn. This left it as nothing on either pile and the imperials won.
This strategy seems unbeatable. We found this mission to be quite frustrating/impossible and it seems the only way to counter is to not place any barrels down so they can't be stolen until the last mission which doesn't seem very flavor of the mission and is quite boring.
How did you guys read the 3 dmg thing?
3 damage suffered In a single attack. See suffers vs. has suffered from the RRG. There is nothing in the middle unless the rules are explicit about it.
Haven't read the mission yet, but it sounds kind of like CTF in Imperial Assault. I'm intrigued. Whole lot better than Viper's Den, I hope.

Why it cannot be Non-Sentient figure ? That figure cannot perform "use" action. During this mission you don't make "use" action like in case with doors, crafts or terminals, you just pick up for your movement points. So Nexu also can take barrel in my opinion.
Retrieve contains an interact whether you need to spend an action or not. There's also an existing ruling confirming that.

Because Non-Sentient prevents interact, it also prevents retrieve.

I feel like such an idiot. I was googling where the Neutral Tokens were on the map because I thought the deployment section was just figures. Lol. But finding this, I do like all the comments and strategies.
