Nope. Presumably you're talking about Assets, like Firearms or First Aid. Those say "Uses X", meaning when you put them into play, place X resources on them from the token pool. Those are the Supply that you spend in order to use that card's action. Once those supplies are gone, you can't use it anymore.

This is correct.

However, for clarity, even though the card can not be used, unless it says otherwise, it stays in play.

The ones that say otherwise are the exception (First Aid, Forbidden Knowledge and Grotesque Statue). 2 of those (First Aid and Forbidden Knowledge) are slotless, while all of those that stay in play take one or more slots. So you can keep a used up asset (in case you fail the test for Crypt Chill, for instance), at least until you have another card you want to put in that slot.

what made you think the supply was in your hand to begin with?
When I was using "Flashlight", I thought the (3 Supplies) meant I can only use the cards effect three times. Then I thought I had to spend the supply in my hand, which would make the game much harder. I didn't question it because it's Arkham and I wouldn't put it past the game.
