Had a chance to play AH:LCG last night at a pre-release party last night. Really enjoyed the game! However all four players discovered we were missing Encounter Card #14 out of the 16 card set. I was wondering if anyone else has experienced this issue.

It's Lita Chantler. The card is not missing, but it's included out of number order.

Several players had reported that this card is "missing", but found it after looking for it again.

And it's out of number order because it has a player deck back, instead of an encounter deck back. Most likely it is where it is in the packaging due to bundling restraints at the factory.

Is this a misprint or by design?

By design. Lita is never shuffled into the encounter deck (she's set aside at the start of the first scenario) and you might get to add her to a player's deck, depending on the resolution you reach. So she has a player back because that's the kind of deck she's going to be shuffled in.
