So the new timing chart in the FAQ says that attack dice (and defense dice) can be modified multiple times but each dice can only be rerolled once. Does this mean that if I attack with Han and I have a target lock, I have to choose between either using Han's ability or spending the target lock to reroll dice?

You wouldn't want to most of the time but it is conceivable. Suppose you have Blinded Pilot so can't shoot this round... but also nobody can shoot YOU. Your action might be target lock. Suppose you got a focus from Kyle Katarn? Suppose you roll CRIT CRIT BLANK BLANK at range 1 and don't want to use Han's ability, but rather the TL on the blanks?

Crabbok has it right. It won't always happen, but if there are no other actions that would be beneficial then you may as well. It persists between turns, and lets you reroll some dice without rerolling all of them.

It happens very rarely. If you get into range of Palob's ability and can't/don't boost.

Why would you EVER target lock with Han?
1. For some crazy reason you are using ordnance that requires a TL to use.
2. You are using some other upgrade that utilizes the TL mechanic.
3. You have a situation where you do not wish to reroll ALL of your dice. TL allows you to choose which to reroll while Han just say "pick'em up and try again."
I've used Han w/ ptl, kyle, and kanan (along with engine and title) quite extensively and in that build I use TL all the time, and Han himself is more of a PS bid than anything.

TL still has some benefits over Han's ability. With Han you have to reroll all dice including the hits and crits. With TL you can keep the hits and crits. Say you got 1 crit and all blanks using the TL would be preferable than Han's ability.
As for modifying dice any number of times so say if shooting with mangler cannon you can reroll you dice change rerolled focus into hits with a focus token, and even change a hit to a crit with mangler.

Because when you use Han's ability there's absolutely no guarantee that you'll get a better roll. In fact, as it often happens, you may roll equal or worse.
However, with Target Lock, you won't roll worse. The roll will be at worse equal, often better.
