Can a Hero shoot through other Hero's to a target or do they block LOS to target?

No, you can't shoot through other heroes. All figures block LOS for all attacks, unless you have a hero ability, skill or other card effect that says otherwise. Note that, in one of many curious twists, the villager tokens included in Road to Legend are considered figures for rules purposes. Unlike familiars and other token creatures.

Edit: If the heroes in your games are having trouble with other heroes blocking their shots, remember that you can move-attack-move provided you have enough MPs. There's no rule that says you have to do all your movement before or after your attack. The same applies to the OL's monsters.
