Is it mean that no spells will effect the Amulet as well?

No Spell cast at you will affect any Object or Follower or you. Destruction only allows you to remove a card from the board, not from a character.
The card said that no spell will effect you (the character), the "Shatter" cast on an object, am i right?
And for the "Destruction", i ment if i droped the "Amulet" on the board, can i cast the "Destruction" on it? or spells wont effect him?

Amulet blocks all spells, except the Command Spell from affecting you or your belongings. If it only affected you (character), Mesmerism would be fair game, same for Acquisition, making Amulet pretty useless.

I agree with Dam on everything but he didn't answer to this question.
Amulet protects the Character and his belongings, it doesn't protect itself if it is not worn. Surely you can Destroy the Amulet left face up on the board. As Dam correctly pointed out, you can't use Shatter on Objects on the game board, because the target is the Character.

Hey, it's a Magic Object, Warlock has a Quest for it IIRC.

But if you play random, and you don't have that quest, then you can better not keep it with you in the whole game, until you draw ( maybe) that quest
You can better sell it at the alchemist
