I bought Talisman 4th edition and The Reaper expansion last week and have set up a game for my group this weekend (5 of us including me).
Any tips or advice before we start our adventure?

Have plenty of food and block off a few hours. It may take awhile
Decide how you are going to choose characters, decide what kind of ending you want, get ready to come here or BGG for rule questions.
Good advice.
Have a computer, tablet or phone ready and set on these forums - lots of questions arise during the game and it is quicker to search the forums sometimes than read the rulebook! And then there are the questions that are not covered by the rules that you will find here too.

Oh and having the forums on hand is a great shout chemical22. Seems like hitting a road block when it comes to rules will be the only hurdle to overcome. So with that in mind:
Which rule do you wish was more clear/actually present in the rule book?
first time we played we misinterpreted the rules for the characters that always have 1 spell; you can only cast 1 spell in other players turn and only as many spells during your own turn as you start the turn with; also spells must have an effect you can cast them Fx you can cast nullify (target player discards all spells) on yourself (to get rid of the spell and get a new one); but only if you have another spell that nullify can discard (similarly you can only cast healing if the target is below starting life). The wizard was casting 20 spells a turn until we figured it out^^
it also took a few games before we understood that if you dont defeat enemies you dont get to pick any objecs there may be on the same space.
Both rules are actually clearly laid out in the rulebook though:)
About the time; the game can be really long especially when people dont know the cards. if you know the game and just reveal the card, resolve quickly and next player goes its about 1 hour pr game, if you carefully explain every card you're looking at 4-5 hours. Hopefully not an issue for you guys, just be aware that that's likely what you're looking at.

- warlock's quests and portal of power, completing "immediately"
- definition of "value"
- a fight with craft isn't a "battle"

Lets see - if you play with the Assassin character, you may want to read up here first. Also, the poltergeist follower has a lot written about it too. I'm sure there are lots more, but the real complications with most cards come up with very specific scenarios; and more-so when you add additional expansions to the mix.
Chances are, if you are left questioning about something, there is something here in the forums written about it.
