I agree, it's highly likely that they will require Twin Shadows as the minis directly replace those in the expansion.
I would suspect so as well.
I think it's a tossup right now. They could easily build the maps with core set tiles. But I don't know if they will.
FFG doesn't have a spotless record, but I'll give them the benefit of the doubt on this. I won't be surprised if it happens, but I'm not assuming it will until I see it. If they do go that route with Wave 2, I'll change my expectations for the future. It probably won't affect me, because everything I've seen of Wave 2, I'm probably going to get it all. (I might even split a second set of Twin Shadows itself with a friend who will probably want the tiles and Rebel figures, while I grab the Imperial/Mercenary figures, but that's mostly unrelated to the topic at hand.)
My experience with FFG is it will be a mix of core set and Twin Shadows "deluxe expansion" tiles. Looking at LOTR LCG as a basis they have a deluxe with 2-3 encounter sets in it. Then each time a pack comes out before the next deluxe the stories in the pack contain one new encounter set and then use sets from the last deluxe and sometime a core.
If we see Leia and Dash next I'd expect their skirmish maps to use tiles from the next deluxe and core, but I doubt Skirmish maps will ever use tiles from two different deluxe expansions.
How can these skirmish maps be used in organised play?
The current rules say everyone has to bring their own map tiles, that means that either these maps will never see tournament play or only players who have bought the expansion can take part.

The OP rules already cover this. They list which maps can be used in a premier level tournament. That means any sort of official event, like a store championship, regionals, national or worlds will only be using the skirmish maps listed in the tournament book.
Starting March 1st, the legal maps, and their accompanying missions, for the season are: Mos Eisley Outskirts, Massassi Ruins, and Moisture Farm.
That means in the next season you may see the maps from Twin Shadows included in that list, or you may not. If they do include them, then that means people playing in skirmish tournaments may need to buy Twin Shadows. That assumes that skirmish missions coming with those expansions use the tiles from Twin Shadows.
