If a character is prone it means they're on the ground. Shooters will have a harder time hitting them, so add a setback die to shooting. Melee will have an easier time, so add a boost die. It takes a Maneuver to get up, unless you have the Jump Up Talent.

There is rarely a "supine" condition in RPGs. "Prone" is usually used to mean "lying on the ground," no matter what position you're in. Orjo is correct, though that in ordinary speech "prone" specifically means lying on your stomach and "supine" lying on your back.
The closest thing I've ever seen to a distinction in an RPG is in Fantasy Craft, a d20 offshoot that used "prone" to mean "deliberately lying on the ground, for the sake of concealment or whatever" and "sprawled" to mean "knocked down involuntarily," with different mechanical effects for each. Still didn't use "supine," though.
