My question is how long do the requirements for a combo E have to be in the card poo for them to be valid. That is to say if I play a mid attack and then play Dimonic Self Sacrifice Stratagen and finally play Twilight Embrace. Am I able to use the continuous ability on DSSS to remove itself from the cardpool and still use the combo ability or does DSSS have to stay in the cardpool for until after I have played the Combo E?

The combo has to be "true" when you use it. So DSSS wouldn't work.

However if you had the "combo" and DSSS was messing it up then your scenario could work.
