Let me put it another way: is there anything that prevents putting Luke and Wedge on the same ship?

Nothing at this time no.

My only clue so far is that the Imperial 181st TIE Interceptor specifically says "Its Pilot enhancement." The fact that 'enhancement' is singular leads me to believe a) you can only have one Pilot enhancement per vehicle, b) if you have two on there, you have to choose one.

As Toqtamish said, we don't know yet, and there's nothing currently preventing Wedge and Luke on the same vehicle (which I've done before), but the wording of the above card leads me to believe this will change soon.
